After shutting down and then trying to restart a WS2008 instance i receive a VERR_NOT_SUPPORTED error. Machine was working fine until this issue.

Please could you check if the image file is still accessible for reading and writing from the host? I want to make sure that there isn't a problem on the host (which host is that, btw)?
And please attach the VBox.log file of your last session.
